Donamem (5+10) mg is a drug that contain Donepezil and Memantine as the active ingredients. The combination is mainly used to treat Alzheimer in which the patient brain cell and their connection starts degenerating as a result loss of memory and other mental function loss occurs.
What is Donamen used for?
Alzheimer is the most common form of dementia, a general term used for memory loss and loss of other intellectual abilities. Donamem (5+10)mg is postulated to exert its therapeutic effect by enhancing cholinergic function. This is accomplished by increasing the concentration of acetylcholine through reversible inhibition of its hydrolysis by acetylcholinesterase. If this proposed mechanism of action is correct, donepezil’s effect may lessen as the disease process advances and fewer cholinergic neurons remain functionally intact.
How does Donamem works?
Donamem (5+10)mg is a centrally acting reversible acetyl cholinesterase inhibitor. Its main therapeutic use is in the treatment of Alzheimer's disease where it is used to increase cortical acetylcholine. An early pathophysiological feature of Alzheimer's disease that is associated with memory loss and cognitive deficits is a deficiency of acetylcholine as a result of selective loss of cholinergic neurons in the cerebral cortex, nucleus basalis, and hippocampus. Donepezil is postulated to exert its therapeutic effect by enhancing cholinergic function. This is accomplished by increasing the concentration of acetylcholine through reversible inhibition of its hydrolysis by acetylcholinesterase. If this proposed mechanism of action is correct, donepezil's effect may lessen as the disease progresses and fewer cholinergic neurons remain functionally intact. There is no evidence that Donepezil alters the course of the underlying dementing process.
Memantine also has antagonistic activity at the type 3 serotonergic (5-HT3) receptor with a potency that is similar to that at the NMDA receptor, and lower antagonistic activity at the nicotinic acetylcholine receptor. This drug has no affinity for γ-aminobutyric acid (GABA), benzodiazepine, dopamine, adrenergic, histamine, or glycine receptors or for voltage-dependent calcium, sodium, or potassium channels.
What are the side effects of Donamem (5+10)mg?
There are several side effects of this drug which are as follows fatigue, vomiting, muscle cramps, weight loss, anorexia, and ecchymosis, diarrhoea, loss of appetite, muscle cramps, nausea, trouble in sleeping, unusual tiredness or weakness vomiting, abnormal dreams, constipation, dizziness, drowsiness fainting, frequent urination, headache, joint pain, stiffness or swelling, mental depression, pain unusual bleeding or bruising weight loss.

When should Donamem should not be used?
You should not use Donamem if you are
- Do not drive or use any machinery because using these tablets may have a minor or moderate effect on your ability to drive and using of machines. They may cause dizziness, so avoid driving and use of machines, while taking this drug
- Avoid alcohol while taking this drug
- Donepezil may cause a condition that affects the heart rhythm (QT prolongation). QT prolongation can rarely cause serious (rarely fatal) fast/irregular heartbeat and other symptoms (such as severe dizziness, fainting).
- Low levels of potassium or magnesium in the blood may also increase your risk of QT prolongation.
- Avoid this drug if you are pregnant or a lactating mother
